Bud rot. Get rid asap, 3-5 cm up and down from spot goes to bin too. Check other plants , if its spreading, cut not only this plant, but the rest too. Take your early loses.

Hi and I‘am so sad to tell you: It really looks like bud rot (Botrytis) starting to set in. The brownish/mushy spots inside the cola and the sugar leaves dying off from the bud are strong indicators. Nutrient issues usually show up more evenly on the fan leaves, but here the damage is very localized inside the flower, which is typical for mold. Best thing to do now: Cut out all infected buds immediately (including some healthy-looking material around them). Never smoke or process the affected parts it’s not safe. Improve airflow around the plant and reduce humidity as much as possible. If outdoors, try to protect from evening moisture and rain. Sadly, once bud rot appears, you can’t reverse it you can only stop it from spreading further

That looks like something has eaten/damaged the stalk, cutting nutrients and water to that area, resulting in this damage. Don't paint, but do pinch it out to the stalk (there may be a small worm in the stem, so kill that with a piece of wire or similar) before it moulds and spreads more damage. And it will mould the next time it rains! Hope that helps, it looks like a nice herb!
